For research collaborators

A shared analytical language for your research team.

FRAGSIGN, powered by SDF-Index, supports laboratory research into sperm DNA fragmentation. This overview helps researchers and clinician-scientists understand the workflow and its analytical outputs.

Research measurements

Keep the measurement tied to the method.

DFI and HDS describe different features of the acquired fluorescence data under the assay conditions. Interpretation begins with the protocol, specimen preparation, acquisition quality, and analysis settings.

A consistent report structure supports comparison and review within a research project. It does not by itself establish the validity of a scientific conclusion.

Laboratory context
  • Research protocol
  • Specimen preparation
  • Acquisition conditions
  • Quality-control review
Analytical output
  • DNA Fragmentation Index (DFI)
  • High DNA Stainability (HDS)
  • Research categories
  • Structured report

Understanding the output

Two separate analytical parameters

DNA Fragmentation Index (DFI)

The proportion of measured sperm events exhibiting increased susceptibility consistent with fragmented DNA under the assay conditions.

DFI is reported as a percentage. Research category assignment should be reviewed with its defined boundaries and method context.

High DNA Stainability (HDS)

The proportion of measured sperm events exhibiting high DNA stainability under the assay conditions.

HDS is reported separately from DFI and should remain a distinct parameter in research analysis.

Study review

Ask what the data can support.

Research teams should consider repeatability, acquisition conditions, quality controls, and the relationship between technical replicates and independent specimens.

Useful review questions

  • Which protocol and instrument conditions were evaluated?
  • How were quality issues handled?
  • How do results vary across time and specimens?
  • Which claims are supported by the study design?
